Doing an innoculation from a multispore syringe into bottles of grain. It seems the two major variables are temperature and gas exchange. For temperature I'm not controlling it, relying on ambient temperature. If it is slightly cool and the colonisation process is retarded, will that adversely (or positively) affect the later fruiting stages, or are all fully colonised bottles equal no matter how long it took to fully colonise. As to the gas exchange I'm very confused. I've seen people making big holes and stuffing it with polyfill and self-healing injection ports. Seen people make four holes or one hole sometimes covered with micropore tape sometimes not. I've even heard no lid is required you can use two layers of tinfoil and inject through the bottom layer. The advice out there is very varied. What and how much are the mycelium breathing in and exhaling at this beginning stage; and how do I optimise the gaseous environment in a grain spawn bottle. The last passing thought is the size of the bottle itself. Big or small, or is it a non-issue. Like i said basic questions, more just me showing off my ignorance and inexperience, but any comments to set me straight before i do my next set of innoculations would be appreciated. Thanks

Thanks sladds, I'll give the book a read. Although I've read a bit, got the basic ideas, suppose i was looking for a short cut to circumvent practical experience when that is never possible. . Yes i realise ms isnt the ideal way to do it, but agar work is a little beyond me at the moment. I came into this thinking it would be a simple thing and after scratching the surface and seeing the depth of the subject I've had to revise my thinking.
