Joey Green Your time, insight and wisdom once again, very mush appreciated. I have yet to work on slants but I’ll start looking for the best teks to get started. Unless of course, you wouldn’t mind teaching an eager student. I want to get fruit like this when I grow up, Sir! Who else agrees?!

I can say with certainty that a determining mindset will win 11 times out of 10! Lol Slants sre easy ..... A few years back s scored a couple hundred 50ml pyrex culture bottles with a septa port in the lid on ebay for a steal of a price.... Once sterilized, I simply inject a drop or two of whatever Im working in through the septa port without even having to open the slant. How I prep my slants is to prepare my agar as low in nutrition as possible while still allowing in to gel once cooled. I buy some hardwood popsicle sticks (hardwood is important, but I think most are anyway) .... I cross cut the popsicle sticks in half, making them the perfect height for my slant receptacles .... and then snap these in half long ways (I end up with 4 slivers per stick), put them in my slant jars, pour the proper amount of agar in, loosely put on the caps, load all my "loaded" slants into an autoclavable tube rack, wrap it up with aluminum foil so its like a nicely packaged "brick" and put in pressure cooker for maybe half hour. Upon removing them once sterilized, Ill do so in front of my LFH and place these "bricks" on an angle to cool ...... once cooled I remove the foil, tighten the lids ...... and away we go! Lol Ill usually do maybe 50 or 60 at a time, use what I need, and then store the excess in the fridge until needed ... super duper convenient! They will keep for a surprisingly long time refrigerated and sealed tightly. I like to pour enough agar in so that when I tilt them for cooling/hardening, the agar almost reaches the lip of the culture tube and covers maybe 80% of the popsicle stick ..... this makes for easy removal of mycelium pieces when I do harvest a piece. If harvesting the entire culture, I take sterile tweezers, grab the tip of the popsicle stick and pull the whole thing out. The popsicle sticks are dual purpose .... they aid in long term storage as well as give the mycelium more surface area to attach to. Its much easier than it may at first sound. Finding proper culture slant tubes can be an experiment ..... I have quickly found that shoulderless tubes with a wide opening to be the most convenient and efficient ..... My bottles are 50 ml and maybe 2.5 - 3 inches long and approximately 30mm wide (or slightly larger than a US quarter. Any tube will do .... but planning for convenience and ease of use is always helpful.
